When analyzing intact mAbs using a widepore reversed phase column we would recommend you begin with a short high aqueous hold of around 10-15% 0.1% TFA in water. A typical gradient would then run up to around 40-50% of 0.1% TFA in ACN at 1-2% of B per minute. You should then ramp your gradient to 95% B over 2-5 minutes to clean off the column before re-equilibrating your column at the initial conditions.
