Why is it necessary to optimize the generic method?

The generic method should serve as a starting point of method development.  Below is a list of reasons why a generic method should be optimized:

 

  • Current SPE media volume may need to be optimized for the volume of sample that is being analyzed.  The sample volume could be overloading or underloading the sorbent mass available in the selected SPE cartridge.  Both lead to method inconsistency.  Increasing sample volume for underloaded methods can also save cost
  • Generic wash is too gentle and does not remove majority of the matrix interference
  • There may be recovery loss in the wash if the generic wash is too strong for certain hydrophilic analytes
  • Generic elution may recover both interferences and analytes of interest.  An optimized elution should elute mostly target compounds, leaving interferences in the cartridge.  In this case, it may mean that the wash needs to be slightly stronger to remove interferences prior to the elution
  • Generic elution is not strong enough to elute target analytes and initial results show low recoveries
  • The time and effort spent doing SPE is not maximized unless the method is optimized
  • To improve the robustness of the method over time
